反硝化聚磷菌N14原生质体的制备与再生条件研究

摘要:以反硝化聚磷菌N14为对象,应用原生质体技术,从菌龄,氨苄青霉素钠预处理的浓度、时间,溶菌酶的浓度、酶解时间和温度等方面研究了该菌原生质体形成和再生的最佳条件。结果表明,反硝化聚磷菌N14原生质体最大制备率产生的条件:在菌龄16 h时加入120μg/m L氨苄青霉素钠预处理6 h,1μg/m L的溶菌酶38℃酶解45 min,再生培养基培养48 h后,该菌原生质体制备率与再生率分别为96.7%和40.9%。溶菌酶酶解时间对菌体再生率的影响大于制备率。

Abstract:Using protoplast technology,the best conditions were investigated for the protoplast formation and regeneration about denitrifying phosphorus accumulating bacteria N14,by the fungus age,pretreatment of ampicillin sodium concentration,time,the concentration of lysozyme,enzymatic hydrolysis time,and temperature. The results showed that ampicillin sodium with 120 μg / m L were inserted into the bacterial suspension for6 h when the cultures were trained 28 h. At 38 degrees centigrade,lysozyme with 1 μg / m L enzymolysised cell walls would take 45 min. And then the cultures were trained for 48 h in solid regenerative medium. The rate of preparation and regeneration of N14 were 96. 7% and 40. 9%,respectively. The influence of enzymolysis time to regeneration rate is greater than that to preparation.
